G-quadruplex formation of oligonucleotides containing ALS and FTD related GGGGCC repeat

摘要: A largely increased number of GGGGCC repeats located in the non-coding region of gene have been identified as the leading cause of two related neurological disorders, familial amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). We examined G-quadruplex forming ability of GGGGCC-repeat containing oligonucleotides with four guanine tracts chosen as the smallest possible model to form a unimolecular G-quadruplex. These oligonucleotides are readily to folded into G-quadruplexes in the presence of K ions. However, the formation of multiple structures makes structural analysis challenging and time consuming. We observed that flanking sequences on 5'- and 3'-ends as well as mutations of loop residues have a profound effect on folding. Sequence d[(G C ) G ] was chosen for further scrutiny and optimization of nuclear magnetic resonance (NMR) spectroscopic properties with dG to 8Br-dG substitutions at specific positions in the sequence under different folding conditions. Expectedly, folding into desired predominant topology is facilitated when substituted residue adopted a conformation in the naturally-occurring structure. Single dG to 8Br-dG substitution at position 21 and fine tuning of folding conditions facilitate folding of d[(G C ) GG GG] into (mostly) a single G-quadruplex, and thus enable determination of its high-resolution structure by high-field NMR.

5′-tiRNA-Gln inhibits hepatocellular carcinoma progression by repressing translation through the interaction with eukaryotic initiation factor 4A-I

摘要: tRNA-derived small RNAs (tsRNAs) are novel non-coding RNAs that are involved in the occurrence and progression of diverse diseases. However, their exact presence and function in hepatocellular carcinoma (HCC) remain unclear. Here, differentially expressed tsRNAs in HCC were profiled. A novel tsRNA, tRNAGln-TTG derived 5′-tiRNA-Gln, is significantly downregulated, and its expression level is correlated with progression in patients. In HCC cells, 5′-tiRNA-Gln overexpression impaired the proliferation, migration, and invasion in vitro and in vivo, while 5′-tiRNA-Gln knockdown yielded opposite results. 5′-tiRNA-Gln exerted its function by binding eukaryotic initiation factor 4A-I (EIF4A1), which unwinds complex RNA secondary structures during translation initiation, causing the partial inhibition of translation. The suppressed downregulated proteins include ARAF, MEK1/2 and STAT3, causing the impaired signaling pathway related to HCC progression. Furthermore, based on the construction of a mutant 5′-tiRNA-Gln, the sequence of forming intramolecular G-quadruplex structure is crucial for 5′-tiRNA-Gln to strongly bind EIF4A1 and repress translation. Clinically, 5′-tiRNA-Gln expression level is negatively correlated with ARAF, MEK1/2, and STAT3 in HCC tissues. Collectively, these findings reveal that 5′-tiRNA-Gln interacts with EIF4A1 to reduce related mRNA binding through the intramolecular G-quadruplex structure, and this process partially inhibits translation and HCC progression.

摘要: Poly(2-hydroxyethylmethacrylate) chains were grafted onto the backbone of agar using a microwave assisted method involving a combination of microwave irradiation and ceric ammonium nitrate to initiate the grafting reaction. The synthesized graft copolymers were characterized by intrinsic viscosity measurements, Fourier transform infrared spectroscopy, elemental analysis (C, H, N, O and S) and scanning electron microscopy. Ag-g-P(HEMA)-2 showed a much higher flocculation efficacy than agar. The optimized dosage of flocculation for Ag-g-P(HEMA)-2 in the wastewater was found to be 0.75 ppm. Compared to agar, Ag-g-P(HEMA)-2 was found to considerably reduce the pollutant load in the wastewater.

摘要:

Egg white proteins are one of the major allergens. The objective of this study was to investigate the effect of Aspergillus oryzae cultivation on IgE binding ability of egg white proteins. Effect of A. oryzae on egg white proteins was determined using ninhydrin method, SDS-PAGE, ELISA, fluorescence FITC labeling, MALDI-TOF-MS and LC-MS/MS analysis. Adding mycelium of A. oryzae ATCC 1011 and 16868 substantially reduced the IgE binding ability of acidified egg white after 24 h incubation. The binding capacity of egg white proteins to IgE in plasma from four egg allergy patients was almost completely lost after incubation with mycelium of ATCC 16868. Results from SDS-PAGE, free amino acid analysis, MALDI-TOF-MS and LC-MS/MS indicated that there was no substantial protein degradation during incubation. Therefore, the reduction of IgE binding ability of egg white proteins during A. oryzae treatment was probably due to a loss of ~1700 Da mass including a fragment of the ovomucoid N terminus.

Biogas and its opportunities—A review

摘要: Biogas production is a well-established technology primarily for the generation of renewable energy and also for the valorization of organic residues. Biogas is the end product of a biological mediated process, the so called anaerobic digestion, in which different microorganisms, follow diverse metabolic pathways to decompose the organic matter. The process has been known since ancient times and was widely applied at domestic households providing heat and power for hundreds of years. Nowadays, the biogas sector is rapidly growing and novel achievements create the foundation for constituting biogas plants as advanced bioenergy factories. In this context, the biogas plants are the basis of a circular economy concept targeting nutrients recycling, reduction of greenhouse gas emissions and biorefinery purposes. This review summarizes the current state-of-the-art and presents future perspectives related to the anaerobic digestion process for biogas production. Moreover, a historical retrospective of biogas sector from the early years of its development till its recent advancements gives an outlook of the opportunities that are opening up for process optimisation.
